Cells and cell-free solutions of the culture filtrate of the bacterial symbiont, Xenorhabdus nematophila taken from the entomopathogenic nematode Steinernema carpocapsae in aqueous broth suspensions were lethal to larvae of the diamondback moth Plutella xylostella. Their application on leaves of Chinese cabbage indicated that the cells can penetrate into the insects in the absence of the nematode vector. Cell-free solutions containing metabolites were also proved as effective as bacterial cells suspension. The application of aqueous suspensions of cells of X. nematophila or solutions containing its toxic metabolites to the leaves represents a possible new strategy for controlling insect pests on foliage.

The photodynamic therapy (PDT) is a combination of using a photosensitizer agent, light and oxygen that can cause oxidative cellular damage. This technique is applied in several cases, including for microbial control. The most extensively studied light sources for this purpose are lasers and LED-based systems. Few studies treat alternative light sources based PDT. Sources which present flexibility, portability and economic advantages are of great interest. In this study, we evaluated the in vitro feasibility for the use of chemiluminescence as a PDT light source to induce Staphylococcus aureus reduction. The Photogem (R) concentration varied from 0 to 75 mu g/ml and the illumination time varied from 60 min to 240 min. The long exposure time was necessary due to the low irradiance achieved with chemiluminescence reaction at mu W/cm(2) level. The results demonstrated an effective microbial reduction of around 98% for the highest photosensitizer concentration and light dose. These data suggest the potential use of chemiluminescence as a light source for PDT microbial control, with advantages in terms of flexibility, when compared with conventional sources. Published information on the incidence of pathogens in the field and laboratory infections of Hypsipyla spp. with entomopathogens is reviewed. In addition, some preliminary results of field collections from Ghana and Costa Rica are presented. Fungal pathogens from the Deuteromycetes have been isolated from both H. robusta Moore and H. grandella Zeller. Mermithid nematodes, Hexamermis spp., have been frequently isolated from larvae in the field and incidence of infection with these pathogens can reach significant levels. Microsporidia have been found in cadavers of larvae collected in the field but none have been identified so far. A number of pathogens of other Lepidoptera have been shown to be infectious to H. grandella , including Bacillus thuringiensis , Deuteromycete fungi and a nucleopolyhedrovirus (NPV) from Autographa californica . Hypsipyla spp. are difficult targets for microbial control, since the larvae are cryptic, occur at low density and occur sporadically. In addition, there is a low damage threshold, the plant is susceptible for a number of years and the susceptible part of the plant will rapidly outgrow any surface application. Key features of the biology of entomopathogens with relevance to the control of low density and cryptic pests are discussed. In the light of this experience, we discuss strategies to improve the possibilities of microbial control of this pest and suggest areas for research.

The painted apple moth (PAM), Teia anartoides (Walker) (Lepidoptera: Lymantriidae) made a recent incursion into New Zealand. A nucleopolyhedrovirus (NPV), Orgyia anartoides NPV (OranNPV), originally isolated from PAM in Australia, was tested for its pathogenicity to PAM and a range of non-target insect species found in New Zealand, to evaluate its suitability as a microbial control for this insect invader. Dosage-mortality tests showed that OranNPV was highly pathogenic to PAM larvae; mean LT50 values for third instars ranged from 17.9 to 8.1 days for doses from 102 to 105 polyhedral inclusion bodies/larva, respectively. The cause of death in infected insects was confirmed as OranNPV. Molecular analysis established that OranNPV can be identified by PCR and restriction digestion, and this process complemented microscopic examination of infected larvae. No lymantriid species occur in New Zealand; however, the virus had no significant effects on species from five other lepidopteran families (Noctuidae, Tortricidae, Geometridae, Nymphalidae and Plutellidae) or on adult honeybees. Thus, all indications from this initial investigation are that OranNPV would be an important tool in the control of PAM in a future incursion of this species into New Zealand.

O controlo microbiológico de formulações injectáveis visa garantir a isenção de um produto farmacêutico em termos microrganismos, possivelmente patogénicos, que possam induzir sintomas patológicos após a sua administração. Assim, o estudo das técnicas que permitam a eliminação de agentes patogénicos e a sua prevenção é de extrema importância. Nesta área são utilizadas diversas técnicas clássicas para avaliar determinados medicamentos e estão a ser desenvolvidas outras novas técnicas. Estas últimas revelam-se mais vantajosas comparativamente às clássicas uma vez que apresentam maior eficácia e rigor. Este artigo de revisão apresenta uma descrição simples das principais técnicas disponíveis (clássicas e as mais recentes) para o controlo microbiológico de medicamentos injectáveis podendo ser visto como uma ferramenta prática útil para os profissionais que trabalham ou poderão vir a trabalhar nesta área.

The bacterium from Pseudomonas putida from Steinernema abbasi and its metabolic secretions caused the mortality of the Galleria mellonella pupae. Experiments were conducted in sand and filter paper on time exposure, temperature, moisture, dose and time of penetration of bacterium in pupae and tested stored or dried toxic metabolites using G. mellonella pupae as a test target organism. Death of pupae was probably due to the toxic metabolites. Pseudomonas putida cells were recovered from the haemocoele when bacterial cells were applied to the G. mellonella pupae indicating that bacterial cells can enter the haemocoele in the absence of nematode vector. Penetration of bacterium was found rapidly after application on G. mellonella pupae. Pseudomonas putida or its toxic secretions can be used as a microbial control for insect control. The experimental results indicate that there is possibility of using P. putida and its toxic secretions as a biopesticide and can contribute in the development of new microbial and biological control against insect pests.

Este trabalho teve como objetivo investigar o efeito da temperatura e do teor de umidade do solo na sobrevivência de Metarhizium anisopliae (Metsch.) Sorok. em três tipos de solos. Foram utilizados o Latossolo Vermelho textura argilosa, Latossolo Vermelho textura média e Argissolo Vermelho Amarelo textura arenosa média. As temperaturas empregadas foram 21,5; 26,8 e 31,5°C, e os teores de umidade foram 35, 65 e 100% de saturação. A sobrevivência do fungo foi avaliada após zero, 20, 40, 60, 80, 100 e 120 dias de incubação em cada temperatura estudada. Na análise do efeito do teor de umidade, a sobrevivência foi avaliada após zero, 14, 28, 42, 56, 70, 84, 98 e 112 dias de incubação à temperatura de 27,0±1,0°C. em ambos os ensaios, foi determinado o número de unidades formadores de colônias (UFC) em placa de Petri. Houve influência significativa da temperatura e do teor de umidade na sobrevivência do fungo. O maior crescimento e a maior sobrevivência ocorreram nas temperaturas de 21,5 e 26,8°C, enquanto que, no solo incubado a 31,5°C, o fungo cresceu pouco, e a população declinou rapidamente. No teor de 65% de umidade, houve rápido crescimento do fungo, mas no 112° dia foi observado um declínio da população nos três tipos de solos. Nos teores de 35 e 100% de umidade, o crescimento foi menor, mas obteve-se maior sobrevivência do fungo no solo.

The lace bug, Leptopharsa heveae is an insect that causes serious damage on rubber trees. In laboratory condition, the strains of Verticillium lecanii (ARSEF 6430, 6431 e 6432) and Aphanocladium album (ARSEF 6433) were tested on third-and fifth-instar nymphs and adults of L. heveae to evaluate their virulence using 2.4 x 10(5) and 2.4 x 10(7) conidia/mL. The bioassays were carried out using Petri dishes whose inner bottoms were covered with damp filter papers. Each Petri dish contained five insects and one rubber tree leaflet. The plates were covered with PVC film to provide high relative humidity, maintained at 26 +/- 0.5 degrees C and a photophase of 14 hours. The Probit analysis was calculated from mortality date of insects killed by fungi. In the highest concentrations, ARSEF 6430 was more virulent for third instar nymphs, and the LT50 was 1.9 days. For the fifth instar, the strains ARSEF 6430, 6433 and 6432 showed similar virulence with LT50 of 2.6, 2.6 and 3.2 days, respectively. For adults, ARSEF 6431 was the most virulent strain with the LT50 recorded at 2.0 days. The smallest concentration did not always cause more than 50% mortality.

Bioinseticidas fúngicos são aplicados com pulverizadores convencionais e algumas características destes equipamentos podem não ser adequadas e afetar a eficiência dos bioinseticidas. O objetivo deste trabalho foi verificar se pressões usuais, em aplicações com o pulverizador de barra, podem afetar a ação inseticida de conídios de Beauveria bassiana e Metarhizium anisopliae, componentes de produtos comerciais brasileiros. Para isso, suspensões aquosas de três bioinseticidas foram submetidas à passagem pelo equipamento em três pressões (20, 40 e 60 lbf pol-2) e avaliadas quanto ao rompimento, à viabilidade e à virulência dos conídios. Avaliou-se a viabilidade em lâminas de microscopia cobertas com meio de cultura, após incubação a 26±0,5ºC e fotoperíodo de 12 horas. A concentração foi determinada por meio de contagens em câmara de Neubauer e a virulência foi avaliada para lagartas de Diatraea saccharalis. Não foram encontradas, nos três produtos, influências significativas em nenhum dos parâmetros. Nas pressões avaliadas, a aplicação com o pulverizador de barra não reduz a viabilidade e nem a virulência dos conídios dos bioinseticidas testados, tampouco provoca destruição dos conídios.
